Acupuncture at the Zusanli Acupoint Suppresses Intrastriatal Hemorrhage-induced Increase in c-Fos Expression and Apoptotic Neuronal Cell Death in Rats

Abstract
Intracerebral hemorrhage is one of the most devastating types of stroke. This disease is known to cause severe neurological damage and also has a very high mortality rate. c-Fos is an immediate early gene, and c-Fos expression is sometimes used as a marker for stimulus-induced change in the metabolic activity of neurons. Brain injuries are known to rapidly induce the expression of the c-Fos. c-Fos expression is known to be implicated both in the neuroregeneration and in the neuronal cell death. In the present study, the effect of acupuncture on the c-Fos expression, neuronal cell death, and cell proliferation in the dentate gyrus of rats was investigated via immunohistochemistry for c-Fos, caspase-3, and 5-bromo-2¡¯-deoxyuridine (BrdU). Enhanced c-Fos expression was observed in the dentate gyrus of rats with the intracerebral hemorrhage. Caspase-3 expression and cell proliferation in the dentate gyrus were also increased by intracerebral hemorrhage. Acupunctural treatment, especially at the Zusanli acupoint, suppressed the intracerebral hemorrhage-induced increment in the expressions of c-Fos and caspase-3-, and cell proliferation in the dentate gyrus. The present results suggest that acupunctural treatment may protect neurons from apoptotic cell death and that acupuncture aids recovery from the central nervous system sequelae following stroke.
